Summary:
The purpose of this experiment is to investigate the degradation efficiency of vomitoxin content in corn by different amounts of fungal enzymes. 200g of corn was taken and crushed in a grinder, and then divided into two parts, each weighing 100g; Add 50mg of mycolytic enzyme to one of the two samples, and record as one group; Add another 25mg of mycolytic enzyme and record as 2 groups. 37 ℃ water bath, and samples were taken for testing at 0h, 2h, and 4h. The test results indicate that:
According to the addition of 500mg/kg, the degradation rate of vomitoxin in corn reached 70.02% after 2 hours and 89.38% after 4 hours; According to the addition of 250mg/kg, the degradation rate of vomitoxin in corn reached 51.23% after 2 hours and 78.01% after 4 hours. In summary, the degradation efficiency is best at 500mg/kg, and as the reaction time increases, the degradation efficiency also increases.
1、Materials and Methods
1.1 Test Materials
Fungal hydrolysis enzyme sample: SD BIOTECH GROUP
Corn samples
1.2 Test Method
Sample pretreatment: Take 200g of corn and crush it in a grinder. After crushing, divide it into two parts, each with 100g. Add 50mg of mycolytic enzyme to one part of the two samples, and record it as one group; Add another 25mg of mycolytic enzyme and record as 2 groups; Add aqueous solution to fully dissolve both and stir evenly. Mix the well mixed test solution, check its pH value, and adjust the pH value to neutral. Place the neutral pH value of the test solution in a water bath, and 5. Weigh 5g of the test solution into a 50mL centrifuge tube. Add 25mL of 60% ethanol extract, vortex and mix for 3 minutes, centrifuge at 5000rpm for 5 minutes at room temperature. Take 100 μ L of supernatant into a new centrifuge tube, then add 500 μ L of sample diluent and mix well. Turn on the fluorescence quantitative detector and preheat for 5 minutes. Open the incubator and raise the temperature to 37 ℃. Take 100 μ L of mixed sample and add it to the detection card well. React in the incubator for 8 minutes. Insert the detection card into the detection slot for testing.
